摘要
用高脂血清培养猪主动脉平滑肌细胞,观察HLS对SMC增殖的影响,探讨HLS的作用原理。结果表明:HLS培养能引起类似动脉粥样硬化的动脉SMC增殖。使氚-胸腺嘧啶核苷掺入量增多,形态学上也呈现SMC增生,合成功能增强。而且SMC的增生,随HLS浓度增高而增加。SMC增殖与HLS激发脂质过氧化过程,使培养液中脂质过氧化物升高,前列环素,环磷酸腺苷含量降低,纤溶酶原激活物抑制物活性增高。
The experimental models of porcine aortic smooth muscle cells (SMC) cultured with
hyperlipidemic sera (HLS) were established in order to observe the influence and mechanism of HLS on the proliferation of aortic SMC. The results showed : HLS could faciliate the proliferation of cultured SMC which was close to human atherosclerosis. HLS group had a significantly greater [3H]-thymidine incorporation data than control group with obvious morphological changes of enhanced proliferation and synthesis of SMC. The proliferation of SMC was increased in accordance with the concentration of HLS.
HLS evoked the lipid peroxidation, raised MDA content, plasminogen activator inhibitor (PAD activity and dropped 6-keto-PGF1a, cyclic AMP content. These findings suggest that these model of cultured SMC may be usedful in investgation atherosclerosis, and provide some experimental basis.
