摘要
本实验将编码人TNFa分子上与其受体相关的部分核苷酸序列缺失,代之以人EGF基因cDNA,实现基因重组。转化子经快速细胞破碎法初筛、经Dotblot和Southernblot鉴定,从中挑选出20个人TNFa-EGF阳性重组质粒,为进一步获得重组嵌合人TNFa-EGF分子和TNFa作用机制的深入探讨奠定了初步基础。
In thls study.partial DNA sequence coding the domain of TNFa relating to TNFa receptor was deleted. which is replaced by hu man EG F gene cDNA, So the two genes am fused together. We picked up 20 peresitive humanTNFa-EGF recombinant plasmids with Cracking gel. Dot blot and southern blot,The study lays a primary foundation togetting the recombinant chimeric protein between TNFa and EG F. and to further study of effective mechanism of TNFa.
