雷公藤多甙对小鼠体内姐妹染色单体互换的影响 被引量:4

EFFECT OF TRIPTERYGIUM WILFORDII ON SISTER CHROMATID EXCHANGE IN MOUSE

摘要 雷公藤多甙(GTW)在雄性大鼠及男子均可致不育而无明显毒副作用。本实验用BABL/C小鼠骨髓细胞体内Brdu标记姐妹染色单体互换(SCE)的方法,测定GTW有无潜在的致突变作用。采用临床剂量3倍(3mg/kg)和100倍(100mg/kg),每天灌服,共 7天。另设阳性对照组一次性腹腔注射环磷酰胺30mg/kg。结果,SCE率范围低剂量组在1~7之间,平均值3.63±0.20(SE),高剂量组在1~15之间,平均值 4.56±0.39,阴性对照组在1~8之间,平均值 3.8±0.39,阳性对照组在 19~58之间,平均值33.1±1.06。经统计学处理,低、高剂量与阴性对照组之间无明显差异,而阳性对照组和其他各组之间均有显著性差异(P<0.01)。因而从本实验的结果来看,GTW似无诱变作用。 The present investigation evaluated the possible mutagenic effect of GTW by means of observing the sister chromatid exchange (SCE) in bone marrow Cells of BABL/C mouse.GTW was given orally at dose levels of 3 times (3mg/ Kg.d)and 100 times(100mg/Kg.d)of the clinical dose for 7 days. In addition, recheck control and positive control (Cyclophosphamide 30mg/Kg).were arranged, Results showed that there were no significant difference between the low dose, high dose and the vehcle control groups, but a significant difference (P <0.01) was seen between the positive control group and each of other 3 groups. It is believed that GTWis not mutagenic so far as the results of the present experiment is considered.
