摘要
用鸭源新城疫病毒(ND)D10株和减蛋综合征(EDS—76)病毒GC2株同时在同一鸭胚中复制增殖.结果表明:(1)两种病毒同时感染同一鸭胚后,鸭胚平均死亡时间(MDT)73.6h,死亡率88.2%(15/17).胚体皮肤出血、淤血,绒毛尿囊膜水肿、增厚,尿囊液能凝集人和鸡红细胞.(2)尿囊液感染鸡胚成纤维(CEF)细胞,致细胞病变(CPE),证实尿囊液中存在NDV.用间接法Dot—ELISA检测EDS—76病毒抗原,结果为阳性.(3)电镜观察经提纯浓缩后的尿囊液,可见到两种病毒粒子:NDV形态不一,多数直径约120~130nm或更大.有囊膜;EDS—76病毒呈圆形,直径约70nm,有囊膜.(4)用血凝试验(HA)测定两种病毒.其生长规律与单独接种时一致.(5)接种灭活的含毒尿囊液的雏鸡,在血清中出现抗两种病毒的HI抗体及抗EDS-76病毒的中和抗体,对NDV强毒攻击有坚强的免疫力。
A study was made on the propagation of newcaste disease virus(NDV)duck isolate(Strain D10)and egg drop syndrome 76 (EDS-76)virus(Strain GC2)in one duck embryo.The results were as follows:1.The mean death time of the duck embryos was 73.6 h post inoculation,with a mortality of 15 /17.Gross lesions observed were cutaneous congestion and petechial hemorrhages;edematous chorioallantoic membyane(CAM).The allantoic fluid of infected embryos could agglutinate chicken and human erythrocytes.2.NDV produced CPE in chicken embryo fibroblast(CEF)monolayers,and EDS-76 virus antigens were detected by indirect Dot-ELISA. 3. NDV and EDS-76 virus particles were observed under electron microscope with diameters of 120 ̄130 urn or larger and 70 nm respectively.4.The growth of two viruses coinoculated in one duck embryo was the same as that inoculated separately in two embryos.5.The HI antibodies against tWo viruses and neutralization antibodies against EDS-76 virus were detected in the chicken vaccinated with inactivated viruses. The vaccinated chicken were resistant to the challenge with virulent NDV 20 days after vaccination.
