摘要
猪霉形体性肺炎是一种慢性传染病。其霉形体的毒性及免疫性都与膜浓度和膜蛋白有关。从培养菌中制备足够量的膜制剂是研究该致病原理、免疫机制以及基因表达调控的关键步骤。为此我们在Razin的反复冻融裂解和低渗裂解两种破膜方法的基础上,提出了一种低渗与冻融相结合的新型破膜方法。并通过测定ATP酶和已糖激酶活力以鉴定膜制剂的纯度。试验表明这种方法能提高膜的收获率,达到28%左右。
A mycoplasmal pneumonia of swine is one of the chronic infectious disease. A toxicity and immunity of mycoplasma is closely related to membrane concentration and protein. It is critical process that we prepare a great deal membrane agent from cultivated bacteria, which is very beneficial for us to study a pathogenic principle and immunity and regulation of gene expression of the disease. Having based on that RAZIN' repeated-freezed thaw lytic and osmotic-shock method, we bring up a new split membrane ways that combine repeated freeze-thaw with low-osmotic method. meanwhile, we measured ATPase and hexo kinase in order to appraise purity of membrane agent. The result indicated that the ways can increase gain rate of membrane by 28%.
