摘要
本文采用Iodogen法将鼠抗人胃癌单克隆抗体RSF9(IgM)予以131I标记,研究RSF9在荷人胃癌裸鼠中的定位和显像能力。结果表明,RSF9在体内能特异性地与肿瘤组织结合,T/NT比值随时间延长逐渐升高,96hT/NT比值均大于2.5,瘤/血比值为2.69,肿瘤定位指数达5.18,SPECT得到清晰的肿瘤图像。RSF9具有良好的体内导向定位能力。IgM型单抗也可用于肿瘤显像。
RSF9 monoclinal antibody (IgM) against human carcinoma was labeled with 131I by Iodogen method. The radioimmunoimaging and immunolocalization of 131I-RSF in human gastric carcinoma xenograft were studied. Results showed the xenografts of gastric carcinoma could be visualized clearly by SPECT after intraperitoneal infection of131I-RSF9 at 48, 72 and 96h. Biodistribution study demonstrated that the ratios of T/NT were increasing from 48 h to 96 h;the highest ratio of tumor to blood was 2.96 ±0.45. The localization index (LI) of the tumor was 5.18±0.36 at 96h.The present study indicated that RSF9 McAb may be highly valuable in the guided localization of gastric carcionoma and that the McAb IgM may also be used for radioimmunoimaging.
