摘要 应用MTT比色法,实验研究加温与平阳霉素两者不同顺序的治疗对人食管鳞状细胞癌HEC2的杀伤作用。结果表明:加温和平阳霉素同时处理细胞Lh,细胞杀伤作用最强:先药物处理,1~8h后加温,随时间间隔的延长,其协同杀伤作用逐渐减弱,但强于单独平阳霉素组;先加温,后用药物,时间间隔1~4h,细胞杀伤作用明显减弱,甚至不如单独平阳霉素组,间隔8h,效果高于单独用药组。提示在加温与化疗联合应用中,必须重视热耐受性。 Effect of the sequence of hyperthermia and chemotherapy (pingyangmycin)on human esophageal squamous cell carcinoma celll line HEC2 was examined by MTT assay in vitro.When heat and PYM were applied simultaneously, the cytocidal effect was strongest. When the drug was given 1-8 h before hyperthemia, the cytotoxicity decreased,yet stronger than that of PYM alone. When the heat was given 1-4 h before the drug,the cytotoxicity dropped precipitously,even weaker than that of PYM alone;while in case the interval was extended to 8 h, the cytocidal effect beame stronger than that of PYM alone.It is concluded that the sequence of hyperthermia and PYM influences the cytocidal effect greatly and the best effect was achived when the 2 forms of therapy were applied simultaneously; and care should be taken to guard against the development of thermotolerance when combined hyperthermia and chemotherapy were applied.
