担子菌LMPQ39和CSP菌丝原生质体分离和再生的研究 被引量:1

摘要 本文以蜗牛酶和纤维素酶的混和酶液作为脱壁酶,通过酶解试验比较了菌龄、酶解时间、酶解温度及酶液浓度等对担子菌LMPQ39和CSP菌丝原生质体分离的影响.通过原生质体再生试验,比较了渗透压稳定剂对LMPQ39和CSP菌丝原生质体再生的影响.获得了105~108数量级的原生质体并再生出菌落.同时,本文还对CSP原生质体再生过程的形态变化作了适当的描述. A mixture of commercially available cellulase and snailase released mycelial protoplasts of LMPQ39 and CSP(both belong to basldiomycetes)in yields 105 ̄108/ml mixture.The reaction of enzymatic hydrolysis was going on with mycelial suspentions which were 4 days culture of CSP and & days culture of LMPQ 3 9, utilizing a mixture of 1.0 % snailase 1.0% cellulase at 35℃, with 0. 8mol/1l mannitol as osmotic stabilizer, and reaction mixture within 2. 5h. Protoplasts from LMPQ39 and CSP regenerated hyphae at frequencies of 0. 1%  ̄0. 4% with 0. 6M MgSO4 and 0. 8mol/l mannitol as osmotic stabilizers. Additionally the morphology of the protoplasts regeneration to The mycelial form was also observed by microscope.
