摘要
应用聚合酶链反应、基因克隆和末端终止技术,测定了分属DR9、DR11和DR12的6个中国汉族人HLA纯合细胞DPB1基因第二外显子的核苷酸序列。结果表明,6个纯合细胞分别带有4种等位特异性DPB1*02012、DPB1*0202、DPB1*0501、DPB1*1301,基因结构和白种人相同,因而采用国外参考试剂和探针对中国人标本作DPB1基因分型是可行的。同时,首次揭示了狨猴DPB1基因第二外显子序列,该序列和人类DP基因同源性为69.9%。
By Sanger's chain termination method, the HLA-DPB1 gene second exons of six Chinese HAN consanquineous homozygous cell lines were sequenced. Except that a single strand DNA clone of SMY-23 cell showed a changed code for the 40th amino acid as a somatic mutation,no'new'allele was discovered,indicating a good homology between Chinese and other ethnic groups' DNA samples in terms of nucleotide sequences at least for the four DPB1 alleles, DPB1*02012, DPB1*0202, DPB1*0501and DPB1*1301. In this article. studies on Saoe-DPB1*0101, a marmoset DP allele detected by sequencing the POR amplified segment of cell line B95-8 with the same primers for the HLA-DPB1 second exons was reported for the first time. The SaoeDPB1*0101 shared 58 amino acids with the HLA-DPB1 prototype 0101 in 83 amino acids of first doman of DP beta chain(69. 9%),which revealed an intimate evolutionary relationship between DPB1 genes of human and marmoset Sanguinus oedipus.
