分析人绒毛膜促性腺激素的糖链以诊断滋养细胞肿瘤 被引量:3

ANALYSIS OF THE SUGAR CHAINS OF HUMAN CHORIONIC GONADOTROPIN FOR THE DIAGNOSIS OF TROPHOBLASTIC CELL TUMORS

摘要 用放射免疫法测定正常孕妇(NP)、良性葡萄胎(HM)、侵袭性葡萄胎(IM)和绒毛膜癌(CC)患者晨尿中人绒毛膜促性腺激素(hCG)的含量,发现4组间无显著差别。但用蔓陀罗凝集素(DSA)亲和层析柱分析尿中hCG的结合率,可将良性(HM)和恶性(IM、CC)滋养细胞肿瘤明确地区别开来。HM尿中hCG的DSA结合率与NP无异:而IM与CC尿中hCG的结合率明显高于NP和HM:IM-CC和NP-HM两组间没有重叠。CC尿中hCG的DSA结合率更高于IM尿,但无统计学差别。用小扁豆凝集素(LCA)亲和柱层析则可区分IM和CC的hCG。IM组的hCG的LCA结合率与NP及HM组相似;而CC组的hCG结合率显著降低。以上结果和文献报道的IM和CC中hCG的糖链结构的变化相符合。 Using radio-immuno-assay to determine the human chorionic gonadotropin(hCG) contents in the early morning urine showed no signifieant difference innormal pregnant women(NP) ,patients of hydatidiform mole(HM) ,invasivemole(IM) and choriocarcinoma(CC) .However,benign mole(HM) andmalignant(IM, CC) trophoblastic cell tumors could be clearly differentiated bythe binding rate of urinary hCG on DSA lectin affinity chromatographic column. The DSA binding rate of urinary hCG from HM was the same as that from NP, but the binding rate of hCG from IM and CC were significantly higher than thatfrom NP and HM. There was no overlap between the NP-HM group and IM-CCgroup. The DSA binding rate of hCG from CC was higher,but not significantly,than that from IM.HCG from IM and CC were differentiated by using a LCAleetin affinity column, and CC had a lower binding rate.The above mentioned results were consisted with the structural changes of the sugar chain in hCG of IMand CC patients reported in the literatures
