外源性神经节苷脂对人肝癌培养细胞生长、蛋白分泌及合成的影响 被引量:4

摘要 10mg/L双唾液酸神经节苷脂(GD3)可促进人肝癌SMMC-7721细胞生长,增加细胞DNA合成,静止期细胞数减少而合成期和分裂期细胞增多。单唾液酸神经节苷脂(GM3)对白蛋白的分泌和合成均发生抑制,而GD3对白蛋白的分泌也发生抑制,但细胞内白蛋白的含量上升。GM3、GD3对甲胎蛋白的合成影响不大,GD3可降低甲胎蛋白的分泌,而GM3对甲胎蛋白的分泌影响不大。结果提示GD3有促进细胞生长作用,GM3、GD3比对不同蛋白质的分泌和合成有选择性作用。 At the concentration of 10mg/L, exogenous GD3 could stimulate cell growth of human hepatocartcinoma cell line SMMC-7721, which was most apparent at the fourth and fifth day of cell circle. It was proved through the method of 3H-dT incorporation and cell analysis system that GD3 could increase DNA synthesis and stimulate cell transition from G0/G1 phase to S/M phase.GM3 (10mg/L) could inhibit albumin (ALB) secretion and synthesis; GD3 (10mg/L) could also inhibit ALB secretion, but increase the ALB percentage of cellular proteins. GM3/GD3 had slightly effects on AFP synthesis. GD3 could decrease AFP secretion, but GM3 had no obvious effects on AFP secretion. These results suggested that GD3 could stimulate cell growth, and GM3/GD3 had selective effects on secretion and synthesis of different proteins.
