摘要
胆汁中促抗成核因子的发现是近年来胆石成因研究中的一大进展。我们应用ConA固相化亲和层析结合SephadexG-200分子筛色谱技术,首次从人体胆汁中分离、纯化到一种纯的糖蛋白,表现分子量为子量的此70kD,具有明显的促成核活性,其中性己糖与蛋白质含量之比为15.88%,一分糖蛋白约含640只氨基酸,其N-末端氨基酸顺序为N—Ser—Gly—Met—Arg—Gln—Tyr—Met—Lys。本文同时讨论了此糖蛋对胆石成因的病理学和生物学意义。
n recent years,the big achievement in studying the etiology of cholelithiasis isthe recognition of the role played by the pro and anti-nucleating factors in human bile.Using ConA affinity chromotography combined with Sephadex-200 molecular sieve tech-nique,we have successfully isolated and purified a glycoprotein in gallbladder bile,Ithas obvious pronucleating capicity,and the quantity ratio of neutral hexose to proteinis 15.88%.One molecule of it contains about 640 amino acid residues,The N-terminalamino acid sequence is N-Ser-His Gly-Met-Arg-Cln-Tyr-Tyr-Met-Lys.The pathogenicand biological significance of this glycoprotein in the gallstone pathogenesis have beendiscussed.
