摘要
以水稻的不同遗传型材料的幼穗为外植体诱导愈伤组织,通过继代培养,最终筛选出了来自于品系“145”的胚性愈伤组织.在附加1.5~2.0mg/l2,4—D的NMB固体培养基上继代7次后,转入同种液体培养基上进行悬浮继代培养,由此形成的悬浮系是由单细胞和小细胞团组成的.倒置显微镜下几乎观察不到管形细胞,细胞的成活率达90%以上.未发现酵母抽提物对细胞悬浮培养具有促进作用.在液体培养基中附加600mg/l的肌醇能够大大提高愈伤组织的分散程度,从中分离出的单细胞经初步培养再生了愈伤组织.
Callus was induced from explants of young panicles of different genotypes of rice.Through subculture,embryogenic callus was sifted out from lines'145'The suspension lines obtained through seven subculture on the NMB solid medium and suspension subculture on the same liquid medium were consisted of single cell and small cell aggregates.The tublar cells were not observed under inverted microscope,and the survival rate of cell was over 90%. It was not found that yeast extracts stimulated cell suspension culture.The liquid medium with 600mg/l m-mositol could increase the degree of cell dispersing.The single cell isolated regenerated callus by the priminarily culture.
