脑损伤组织提取液对成年大鼠损伤的大脑皮质神经元的作用 被引量:2

摘要 本研究的目的是探讨神经元的再生。SD成年雄性大白鼠38只,随机配对为实验组和对照组。大鼠经切除双侧大脑皮质顶叶下正中部分(4mm×3mm×1mm),用大约2mm×2mm×1mm海绵胶(gelfoam)粒浸入脑损伤组织提取液(BWTE)中,术时将上述海绵胶粒放入大脑皮质损伤腔内,2个月内加入BWTE4次。对照组用上述方法加入等量的Hnnk’s液,术后60d取材。取含损伤腔的大脑皮质,用Nissl染色,辅以神经丝抗体免疫染色以及电镜结合体现学定量方法分析,结果实验组与对照组相比发现BWTE组可明显促进损伤皮质神经无的存活(P<0.001)。实验组出现较多胞体增大的神经元(P<0.02),且多具有粗大的顶树突。体视学定量方法分析结果是:神经元的线粒体体密度、数密度、面密度以及粗面内质网的面密度均较对照组增加(P<0.05)。提示BWTE对成年大鼠损伤的皮质神经元具有神经营养活性,可能含有神经营养因子(neuronotrophicfactors,NTFs)。 The aim of the study was to investigate the regeneration of neurons. 38 male adult Sprague-Dawley rats were used in this experiment. According to paired design, each of every paired rat was arranged randomly into BWTE treated group and contrlol group. All the rats. under either anaesthesia. were injured by partial decortication bilaterally in the parietal region. The wound,3×3mm in surface area and 1mm in depth, was created with a small spatula. A piece of gelfoam (2×2×1mm3)was soaked with 25μl of BWTE. It was placed in the wound cavity 4 time over a peried of two months.In the control rats Hank's solution was used in stead of BWTE in equal quantity at the same time. 60 days after the operation, the brains were removed out and a 2mm thick block was taken along the coronal plane to inciude most of wound area.The brain tissue was researched quantitatively and qualitatively under light and electron microscope and immunohistochmisiry.
