血小板活化因子刺激肾小球系膜细胞的自分泌作用 被引量:3

摘要 观察了血小板活化因子(PAF)对肾小球系膜细胞(GMC)合成PAF的影响,结果显示PAF能促使OMC释放PAF,且呈剂量效应依赖关系;PAF特异性受体拮抗剂BN52021(BN)可明显抑制PAF刺激GMC产生PAF的能力。提示PAF可活化GMC产生更多的PAF,形成一个自分泌放大环路,这在肾组织损伤中可能起着至关重要的作用。BN可明显地抑制这一作用。因此,本研究为PAF拮抗剂治疗肾脏疾病提供了理论依据。 he effect of platelet-activating factor(PAF)on PAF producted by glomerular mesangial cells(CMC )was investigated, Not only PAF could promote the release of PAF by GMC,but also the results showed a dependence relation between dose and response.BN52021(PAF an antagonist ) markedly inhibited the producton of PAF from GMC after PAF stimulation, The findings indicated that PAF could induce GMC to synthesize more PAF,form an enlargement ring of autocrine. With this effect PAF might play an important role in renal tissue injure.The results also subetantiated that BN52021 could markedly inhibit the effect of PAF,It provided theory evidence for PAF antagonist in treatinent of kidney diseases.
