摘要
本文利用DNA重组技术,将酵母α因子的启动子和信号序列与成熟天花粉蛋白基因融合,从而构建了天花粉蛋白的酵母表达载体。将该载体转入酵母细胞,转化子在选择培养基中培养24小时后,获得了高效表达。表达的天花粉蛋白位于细胞内。
The gene coding for Trichosanthin, a Chinese herb protein was fused to the yeast a factor promoter and its leader sequence. The chimeric gene was constructed in the yeast shuttle plasmid pMM. After transformation of yeast cells with this vector. It was cultured for 24 hours. The highest level of Trichosanthin expressen was detected by anti-Trichosanthin antiserum. The expressed Trichsanthin was found contained in the cell. This is the first report on expression of Trichosanthin in Saccharomyces cerevisiae in the world.
