摘要
用Dig-GST-πcDNA探针分子杂交方法,检测了正常胃组织、胃癌及相应癌旁正常组织中GST-πDNA和GST-πRNA水平.发现GST-πDNA水平没有明显变化,而CST-πRNA在8例胃癌组织中有6例高于正常胃组织,在12例低分比腺癌中有7例癌旁正常组织高于相应癌组织.表明GST-π基因表达增加与胃癌有关,而且早于细胞形态的变化.
Increased expression of the glutathione S-transferase (GST: E, C, 2, 5, 1, 18) π classisoenzyme is associated with both tumor andpreneoplastic tissues. In order to further char-acterize the alteration of GST-π gene expres-sion during progression of carcinoma, boththe levels of GST-π DNA in one normal gas-tric tissue and 20 gastric tumor with perineo-plastic normal tissue. and the GST-π RNA inthe normal gastric tissue and 12 gastric poorlydifferiented adenoearcinomas with correspond-ing perineoplastic normal tissues were testedusing Dig-GST-π cDNA probe by Dot blot hy-bridization. No significant change of GST-πDNA level, but the expression level of GST-πRNA in 6 of eight gastric tumors was higherthan in normal gastric tissue, and in 7 peri-neoplastic normal tissue of twelve poorly dif-ferentiated adenocarcinoma was higher thanthat in its corresponding tumor. This suggeststhat the elevation of GST-π gene expression isrelated to gastric tumor, and earlierthan thechanges of cell morphology.
