CCK-8和NDAP对大鼠脑和脊髓组织c-fos表达的影响

摘要 为探讨八肽胆囊收缩素(CCK-8)和阿片肽相互作用的分子机理,利用抗体免疫沉淀技术研究了CCK-8与NDAP(κ阿片受体激动剂)对大鼠脑(去皮层和小脑)和脊髓背柱组织Fos蛋白的影响。结果表明,0.1μmol/L CCK-8可显著刺激脑和脊髓组织中Fos蛋白增加(分别是对照组的3.8倍和3.6倍)。相同浓度的NDAP对Fos蛋白的生成亦有一定的诱导作用,分别是对照组的2.7倍和2.6倍。 For elucidating the molecular mechanisms underlying the interaction of cholecystokinin octapeptide (CCK-8) and opioids, we investigated the effects and interaction of CCK8 and NDAP (a k opioid receptor agonist) on Fos protein expression in rat brain and spinal cord tissues by in vitro immunoprecipitation method. Under 0. 1 μmol/L CCK-8 stimulation,there were markedly increase of Fos protein levels both in brain (3. 8 fold increase) and in spinal cord(3. 6 fold increase). 0. 1μmol/L NDAP also exhibited inducing effect on Fos protein production, Fos protein level increased 2. 7 and 2. 6 fold respectively in brain and spinal tissues. The combined effect of CCK-8 and NADP on c-fos expression in rat brain was the same as the level induced by CCK-8 alone. However, in spinal cord the Fos protein level induced by CCK-8 combined with NDAP was much higher than that induced by CCK-8 alone.The results indicated that both CCK-8 and NDAP could induce c-fos expression directly, but the patterns of their combined effects on c-fos expression in brain and in spinal cord were different.
