摘要
介绍了一种灵敏的神经生长因子化学发光免疫测定方法。小鼠神经生长因子(mNGF)的抗体IgG用亲和层析纯化并用吖啶酯标记。该法可用于大鼠组织中NGF含量的测定。方法的灵敏度为10pg/mLNGF;批内批间变异系数分别为8.7%及13.2%;回收率平均为103%。mNGF抗体对人的NGF也有极强的交叉反应,故本方法也可能用于病人血清或脑脊液中NGF含量的测定。
A sensitive chemiluminescence assay for nerve growth factor was described. Antibody IgG for mouse nerve growth factor (mNGF) was purified by affinity chromatography and labeled with acridinium ester. NGF contents of rat tissues were determined by this method. The sensitivity of the assay was 10pg/mL NGF. The intra and inter coefficients of variation were 8.7% and 13. 2% respectively. Recovery tests were determined to be 103%. High degree cross reaction between mNGF antibody and human NGF was observed. It may be possible to determine the NGF levels in serum or cerebrospinal fluid of patients by this method.
