摘要
本实验利用细胞培养法和放射免疫测定法研究了TNF-α对hCG诱导离体大鼠黄体细胞孕酮生成的影响及其作用机制。结果发现TNF-α显著抑制hCG诱导离体大鼠黄体细胞孕酮分泌和cAMP生成,具有显著的时间和剂量依赖关系。但TNF-α不影响Forskolin诱导cAMP生成和孕酮分泌,亦不影响PMA诱导孕酮释放。提示TNF-α通过抑制hCG的作用参与黄体机能的调节。
The present study was designed to investigate the effect of TNF-α on progesterone release by using cultured rat luteal cells and to explore its mechanism of action. The results are as follows: 1. TNF-α significantly inhibited hCG- induced progesterone production by rat luteal cells in vitro in a dose and time-dependant manner; 2. Pretreatment with TNF-α(1000u and 3000u/ml) for 24 h significantly suppressed hCG-induced cAMP production. 3. Pretreatment of cultured rat luteal cells with TNF-α for 24 h had no effect on Fsk-induced cAMP production and progesterone release and PMA-induced progesterone release. In brief, TNF-α suppresses progesterone production by cultured rat luteal cells and the effect is due at least in part is due to its suppression of cAMP production.
