Protection role of zinc, selenium, manganese and nickel for the islet βcell trauma

摘要 研究目的探讨Zn、Se、Mn、Ni对胰岛β细胞损伤的保护作用及其机制,为糖尿病的治疗开辟新的途径。研究方法用Wistar大鼠进行体外及体内实验。胰岛细胞体外培养,设正常对照,加Zn孔、加Se孔、加Mn孔、加Ni孔,培养1h测各孔胰岛素含量。然后加入脉链菌素(STZ),再测胰岛素含量。加16.7mM葡萄糖做刺激试验,培养3天,取培养液测脂质过氧化物(LPO)。60只雄性大鼠随机分为6组,即Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ、Ⅵ组。Ⅱ~Ⅳ组腹腔内注射STZ55mg/kg,Ⅰ组仅注同量的构检酸缓冲液。Ⅰ、Ⅱ组喂基础饲料,Ⅲ~Ⅵ组在基础饲料中分别加入ZnCO3、Na2SeO3、MnCl2、NiCl2。72h后出现轻重不等的DM现象,继续饲养3周处死,取胰腺组织做病理检查,胰腺匀浆测血糖、胰岛素、LPO、SOD及谷胱甘肽过氧化物酶(GSH-PX)。结果体外实验结果:加Zn、Se、Mn、Ni培养的胰岛细胞,其胰岛素分泌量、葡萄糖刺激胰岛素分泌量,明显高于单加STZ,而LPO值明显低于单加STZ。体内实验结果:Ⅰ、Ⅲ~Ⅵ组血糖、LPO均明显低于Ⅱ组,胰岛素、SOD、GSH-PX均明显高于Ⅱ组。胰腺病理检查:STZ组大鼠胰腺均有炎症改变几乎见不到正常日细胞,而加Zn、Se、Mn、Ni组大鼠胰岛β细胞破坏较轻。结论Zn、Se、Mn、Ni均对胰岛β细胞有不同程度的保护作用,? Objectives To search for the protection role of zinc (Zn)、selenitim (Se)、manganese (Mn) and nickel (Ni)for the islet β cell trauma and its mechanism, so that to open a new therapcutic method for diabetes.Methods Wistar motise's islet cells were taken up and culfured in vitro. Set up common control pore,adding Zn, Se, Mn and Ni pores.The insulin contents in all of pores were determined after culture of 1h.Then, streptozotocin (STZ)was added into all of the pores, and the insulin contents of all the pores were determined again. 16.7mM glucose was added into the pores so that to do stimulation test, and cultured for 3 days. Then, the culture fluid was take up, and the Lipid peroxide (LPO) was determined. The 60male mice were randomly divided into 6 groups. The groups were respectively called as group Ⅰ、Ⅱ、ⅢⅣ、Ⅴ and Ⅵ. In grotip Ⅱ ~Ⅵ, intraperitoneal injection with STZ 55mg/kg, ingroup Ⅰ, intraperitoneal injection with sodium citrate buffer solution. Group Ⅰ and Ⅱ were fed with basic forage, Basis on basic forage, ZnCO3, Na2SeO3, MnCl2 and NiCl2 were respectively added for group Ⅲ ~Ⅵ, fed them for 5 weeks. Panqreas Pathological examination was carried out, glucose, instilin, LPO, SOD and GSH- PX were determined.Results The results of experiment in vitro: Cultured islet cells adding Zn,Se, Mn and Ni, the insulin secretory volume and glucos3 stimulation insulin secretory volume are higher than that adding STZ, but the LPO value is obviously decreased. The results of experiment in vivo: the glucose, LPO levels are lower and the insulin, SOD and GSH -PX levels are higher in group Ⅰ, Ⅲ ~Ⅵ than that in group Ⅱ. The Pancreas pathological examination show that the trauma of 6 cells is lighter in group Ⅲ ~Ⅵ than that in group Ⅱ.Conclutions Zn,Se, Mn and Ni have protection role in different degree for the islet o cells trauma.Their Protection role may be completed through raising the activity of antioxidase
