Dot-IGSS,Dot-IGS和Dot-ELISA检测脑脊液中循环抗原诊断脑囊虫病的比较研究 被引量:3

摘要 应用自制的直径20nm的胶体金标记GAR-IgG,以斑点免疫金银染色法(Dot-IGSS)、斑点免疫金染色法(Dot-IGS)和Dot-ELISA三种方法,同时检测99例脑囊虫病人脑脊液(CSF)中循环抗原(CAg),阳性率分别为94.95%、93.94%和92.93%。检测51份非囊虫病患者和62例其他脑部疾病患者的CSF,三种方法均为阴性。诊断指数分别为194.95%、193.94%和192.93%。检出CAS的最小量Dot-IGSS为244ng/ml.Dot-IGS和Dot-ELISA为488ng/ml。试验表明三种方法诊断脑囊虫病具有敏感性高、特异性强、重复性好和方法简便易行.有希望用于脑囊虫病的临床诊断。 GAR-IgG was labelled with 20 nm-diametered colloidal gold particles prepared in this laboratory. Detection of circulating antigens (CAg) of cerebrospinal fluid (CSF) by Dot-IGSS, DotImmunogold staining (Dot-IGS) and Dot-ELISA for diagnosis of 99 confirmed cases of cerebral cysticercosis. The positive rates of the Dot-IGSS, Dot-IGS and Dot-ELISA were 94. 95%,93. 94 % and 92. 93% respectively.Detection of CSF from 51 patients with non-cysticercosis and 62 patients with other cerebral diseases were all negative by these three methods. The detection indices were 194. 95%,193.94% and 192.93% respectively. The minimal CAg volume which could be detected was 244 ng/ml by Dot-IGSS and 488 ng/ml by Dot-IGS and Dot-ELISA.The results indicated the assays were highly specific, sensitive. reproducible and promising for the diagnosis of cerebral cysticercosis.
