应用反向高效液相色谱法分析日本血吸虫抗原

摘要 应用反向高效液相色谱法对纯化的日本血吸虫分子抗原进行了氨基酸分析,并比较了等梯度洗脱与线性梯度洗脱的色谱分离效果。结果表明,该法具有快速、灵敏、精确等特点,尤其适用于微量抗原分析。色谱分离效果以线性梯度洗脱为佳。 In this preliminary study,amino acids compsition in the purified antigen from schistosoma japomicum adult worm were determinated with o-phthalaldehyde-β-mercaptoethanol precolumn derivatization,reversed.gradient elution and fluorescence detection,The efficient of separation by the reversed-phage high performance liquid chromatography(HPLG) was compared between the equal gradient elution and linear one.The result showed that the later was better than the former when both were used as separation reagents,and the chromatographic conditions in the present experiment were also briefly discussed.It was considered that the method was a highly sensitive,rapid,accurate and especially suitable in the minimal analysis of amino acid composition for the purified antigens.
