天津地区抗霍乱弧菌单克隆抗体(McAb)细胞系的建立及其应用的研究

摘要 用 SP_2/0小鼠骨髓瘤细胞与 EL-Tor 霍乱弧菌免疫的 Balb/c 小鼠脾细胞,在 PEG-1000作用下进行融合,获得9株分泌抗霍乱弧菌 McAb 的杂交瘤细胞系。用 ELISA 法对其中两株分泌的 McAb 进行特异性、敏感性实验,免疫球蛋白的类型鉴定及染色体检查。并用羟基磷灰石梯度提纯的 McAb 与兰州生研所生产的多克隆抗体(PcAb),对200例临床粪便标本,用双盲法进行快速检测。结果表明,McAb 的敏感性和特异性均优于 PcAb,同时也证实 McAb 应用于霍乱病人的诊断及菌株鉴定的可行性。 Exponentially growing SP_2/O mice mycloma cell were fused with spleen cellsfrom immunized Balb/C mice with EL-Tor vibrio cholerae by the aid of polyethy- lenglycol (PEG)-1000,nine hybridoma cell lines which produced monoclonal antibody(McAb) against vibrio cholerae were isolated.The specificity and sensitivity of twocell lines secreting McAb were detected with enzymelinked immunosorbent assay(ELISA).Immunoglubulic class and chromosome examination were done,McAb thatwas purified with hydroxyapatite by gradient elution and polyclonal antibody (PcAb)produced by Lanzhou Institute of Biological Products were detected on two hundredstool samples with a doubleblind trial.The result showed that McAb was more sen-sitive and specific than PcAb.These findings suggested that the McAb might proveto be useful immunodiagnostic reagents in diarrheal disease and to be capable ofidentifing vibrio cholerae strain.
