摘要 应用〔~3H〕dTdR 掺入肝细胞 DNA 方法测定本室制备的多种动物 HSS 生物活性。断奶大鼠、猪、兔、畸型儿 HSS 刺激 SD 大鼠肝细胞 DNA 合成的活性分别是对照的3.06倍、2.53倍、2.29倍、1.71倍(均 P<0.025),成年猪 HSS 则无差异。显示 HSS 活性产生的年龄依赖性和作用无种属特异性。而对脾、肾、淋巴细胞 DNA 合成无刺激作用,表明 HSS 活性具有器官特异性。〔~3H〕dTdR 体内,外掺入 DNA 两种方法对比研究证实,二者具有良好可比性。 Hepatic Stimulator Substance (HSS) of several weaning animals was determinedwith [~3H] dTdR incorporated into rats liver parenchymal cell DNA.HSS activityof weaning animals including rats,pigs,rabbits and anomaly infants produced res-pectively a 3.06-fold, 2.53-fold,2.20-fold and 1.71-fold increase in the incorporationof [~3h] dTdR into liver DNA of 34%-hepatectomized test animals as compared withcontrol animals using 0.9% NaCl instead of HSS,while in adult pigs it was the sameas the control.The result showed the production of HSS activity was dependentupon age and the effect of HSS was proved to be organ-specific but species nonspe-cific,stimulating the liver but not the kidney,spleen or.lymphocytes.The incorporation of [~3h] dTdR into liver DNA both in vitro and in vivo gavegood comparable results in multiple samples from the same or different animals.
