摘要
分别从三角帆蚌、褶纹冠蚌、背角无齿蚌的外套膜中部分提取了一种碱性磷酸酶(ALP),并对这3种来源的ALP进行了比活力和动力学性质的比较研究。经过相同提纯步骤后,外套膜ALP比活顺序是褶纹冠蚌>背角无齿蚌>三角帆蚌;三角帆蚌ALP作用于底物磷酸苯二钠的最适pH值为9.40,最适温度为37℃,米氏常数Km为0.53mmol/L;褶纹冠蚌ALP的最适pH值为9.12,最适温度为40℃,米氏常数Km为1.72mmol/L;背角无齿蚌ALP的最适pH值为9.50,最适温度为40℃,米氏常数Km为0.57mmol/L。3种蚌的ALP均为Mg ̄(2+)、Ca ̄(2+)、Ba ̄(2+)所激活,为Cu ̄(2+)、Zn ̄(2+)、KH_2PO_4、ME(疏基乙醇)、EDTA-Na_2所抑制。
An alkaline phosphatase was partly purified from the pallium of Hyriopsis cuminigii(Lea),Cristaria plicata (Leach),Anodonta woodiana (Lea)respectivel. The ALPs of different sources were compared under the same conditions.The results are as follows:After the same purification steps,the order of the specific activity is:Cristarta plicata>Anodonta woodiana>Hyriopsis cumingii.The optimal pH and temperature and Km of Hyriopsis cumingil’s ALP for the hydrolysis ofdisodium phenol phosphate are:pH9.40,37℃and 0.5 3mmol/L; the data of Cristaria plicata’sALP are:pH9. 12,40℃and 1.7 1mmol/L;the data of Anodonta woodiana’s ALP are pH9. 50,40℃ and 0.57mmol/L.The ALPs of these mussels are all activated by Mg ̄(2+), Ca ̄(2+) and Ba ̄(2+) but inhibited by Cu ̄(2+),Zn ̄(2+),KH_2PO_4,ME(2-mercaptoethnol)and EDTA-Na_2.
