摘要
铜绿假单胞杆菌PAO1经过亚硝基胍诱变获得了弹性蛋白酶缺失的突变株。经弹性蛋白营养琼脂平板法,从10次独立处理的60000个菌落中得到71株该酶缺失的突变株,其突变株对N-3-氧代己酰-L-高丝氨酸内酯(HSL)诱导物有无反应分成2组。PANO67菌株在加入诱导物后能恢复该酶产生,经过菌落形态、生化特征和细胞膜蛋白质成份的分析,除该酶产生缺失外,突变株PANO67与亲本菌株PAO1相同,结果表明,PANO67是弹性蛋白酶操纵子的调节基因发生突变的诱变菌株。
The paper describes the process of isolating the negative or defective elastase mutants ofPseudmonas aeruginosa PAO1 by means of mutagenesis with nitrosoguanidine as the agent.By way of the e-lastin-nutrient agar plate,71 mutants were obtained from 60 000 colonies. Then the mutants were divided in-to two groups according to each one's response to an autoinducer,N-(3-oxohexanoyl)-L-homoserine lactone.It was found that in the presence of the autoinducer a mutant,PANO 67,could restore elastase production.From the analysis of its biomorphological,biochemical properties and cell membrance fractions,it could beseen that,except for the lack of elastase activity,the mutant PANO67 has the same properties as its parentalstrain PAO 1.The mutant PANO 67 is thus presumed to be a mutant which has a mutation in the regulatorygene of the elastase.
