肿瘤性免疫抑制因子对免疫功能的影响及其理化特性

摘要 本研究观察了几种不同肿瘤细胞株培养上清对ConA诱导的淋巴细胞增殖反应、IL—2产生、IL—2反应性及IL—2受体表达的影响,并对培养上清中肿瘤性免疫抑制因子(TDSF)的理化特性作了初步分析。实验表明:几种不同来源的TDSF对上述免疫功能均有明显的抑制作用,而且TDSF对热、pH均敏感,部分对胰蛋白酶敏感,但反复冻融对TDSF活性无影响。 In this paper, we studied the immunosuppressive factors produced by several histologically distinct tumor cell lines, which were referred to as tumor-derived immunosuppressive factor(s) (TDSF). The TDSF inhibited the mouse spleen cells response to concanavalin A (ConA). They also inhibited the interleukin 2 (IL-2) production . IL- 2 responsiveness and interleukin 2 receptor expression, which may be a major mechanism by which TDSF inhibiting the lymphocyte proliferation and other immune response. However. non-tumor cells did not produce TDSF. These results suggest that TDSF may be one of reasons contributing to the immunodeficiency state detected in the patients bearing tumor. The TDSF were very heat-liable and stable to repeated freezing -thawing. They were sensitive to pH<4. 0 and PH> 11. 0 and resistant to trypsin treatment except the TDSF from YAC - 1 T Lymphoma. The TDSF purified from P815mastocytoma cell supernatant had an M. W of 79KD. The production of TDSF inhibited by mtomycin C indicated that the TDSF were genic products of tumor cells.
