摘要 用荧光偏振技术及孔雀绿-磷钼酸复合物比色法,研究胰岛素对人红细胞膜的流动性及ATP酶活性的影响。结果表明,胰岛素浓度在(0~5)×10^(-9)mol/L时,膜的荧光偏振度及Na^+/K^+-ATP酶活性均随胰岛素浓度的不同而变化,变化的相关性分别为r=0.9998,P<0.05和r=-0.9989,P<0.05。也发现,荧光偏振度和Na^+/K^+-ATP酶活性之间呈负相关r=-0.9984,P<0.05。证明胰岛素与正常人红细胞膜作用后,红细胞膜Na^+/K^+-ATP酶活性的降低与红细胞膜流动性降低有密切关系。 The effects of insulin on human erythrocyte membrane Na+/K+-ATPase and Mg2+-ATPase activities as well as membrane fluidity were investigated by using malachite green-pnosphato-molybdic acid colorimetric method and fluorescence polarization technique. The results showed that changes of fluorescence polarization values and of Na+/K+-ATPase activity were in relation to the changes of insulin concentration within the range of 0-5 10-9mol L (correlation coefficient r = 0.999 8 and -0.9989, respectively). The relationship between the variance of fluorescence polarization values and Na+/K*-ATPase activity was in negative correlation (r = -0.998 4) . The results suggested tnat the decrease of tne Na+/K+-ATPase activity was closely related to the decrease of membrane fluidity with increased fluorescence polarization after interaction of insulin with erythrocyte membrane.
