摘要
将FRTL-5细胞传代后培养,分成4组,分别加入1例Graves氏病、l例甲状腺癌和1例正常人血清IgG(浓度为2g/L)及hTSH(浓度为1U/L),继续培养42小时。经Bouin氏液固定,免疫组化染色显示甲状腺球蛋白。镜下对4组FRTL-5细胞形态进行观察,并对Tg阳性细胞作半定量分析。结果表明Graves氏病者血清IgG有明显促进FRTL-5细胞合成Tg的功能,而甲状腺癌和正常人血清IgG则无促进Tg合成功能,故可将FRTL-5细胞合成Tg的程度作为反映该细胞功能状态的一项形态学重要指标。
A clonal strain of rat thyroid cell (FRTL-5) in continuous culture has been used to investigate the effect of IgG preparations derived from the sera of a patient with Graves' disease and a patient with thyroid carcinoma on the synthesis of thyroglobulin. In the culture medium on the surface of cover glass, we obtained a single layer of the rat thyroid epithelial cells. After fixation by Bouin's solution and immunohistochemical stain (PAP method), we determined quantitatively the thyroglobulin (Tg) positive thyroid cells and discovered that the serum autoimmun-oglobulin of Graves' patient could promote the synthesis of Tg in FRTL-5 cells as the effect of hTSH. But the serum of thyroid carcinoma patient gave the same result as the effect of normal controls.
