摘要
目的:采用毛细管区带电泳法对四物汤及其配伍组方中酚酸类成分——丹皮酚、苯甲酸和没食子酸同时进行含量测定。方法:采用熔融石英毛细管柱(75 μm×49.2 cm,有效柱长39.0 cm)作为分离通道;用30 mmol·L^(-1)硼砂缓冲溶液(体积分数5%甲醇,pH 9.4)为运行缓冲液;运行电压20 kV、检测波长212 nm、毛细管柱温25℃。结果:丹皮酚、苯甲酸和没食子酸分别在3.680—230.0μg·mL^(-1)、1.126-112.6μg·mL^(-1)和3.000—187.5μg·mL^(-1)浓度范围内线性关系良好(r>0.997);回收率在98.6%-102.3%之间;重复性实验中丹皮酚、苯甲酸和没食子酸峰面积的相对标准偏差(RSD)分别为3.99%,4.05%,5.21%(n=6)。结论:测定方法简单、灵敏、快速,回收率和重现性好。
Objective: To develop a method for simultaneous determination of paeonol, benzoic acid and gallic acid in Siwutang and its preparations by capillary zone electrophoresis. Methods: A fused silica capillary (75μm×49.2 cm,with effective length of 39.0 cm) was used. The running buffer was composed of 30 mmol·L^(-1) borax(5% methanol,pH=9. 4). The applied voltage was 20 kV,and the detection wavelength was 212 nm. The capillary temperature was set at 25℃. Results: A good linearity between peak area and the concentration was found in the range of 3.680-230. 0μg·mL^(-1) for paeonol, 1.126-112.6μg·mL^(-1) for benzoic acid and 3.000-187.5μg·mL^(-1) for gallic acid (r>0.997) ,respectively. The recoveries were between 98.6%-102.3% ,the reproducibility (RSD) of this method was 3.99% ,4.05% and 5.21% for paeonol,benzoic acid and gallic acid,respectively(n=6). Conclusion: The method is simple, sensitive and rapid with satisfactory recoveries and reproducibilities.
