摘要
绿脓杆菌是烧伤感染的主要病原菌之一,用淋巴细胞杂交瘤技术制备抗绿脓杆菌单克隆抗体进行保护小鼠抗绿脓杆菌感染实验,获满意效果,再用人体单层中性粒细胞技术,体外检测了抗绿脓杆菌I群10ll5株单克隆抗体(A4和D4)的调理吞噬活性,证明其可显著增强人体中性粒细胞对绿脓杆菌的吞噬,应用于烧伤创面绿脓杆菌感染的治疗,亦获良好疗效。
ybridomas (A4 and D4) secreting mono-clonal antibodies against
pseudomonas aerugi-nosa were produced by fusing the immunizedsplenic lymphocytes with
the mouse myelomacell lines SP2/0. The protective capacity ofthe monoclonal antibodies A4
(IgG 1 ) and D4(IgM) was examined in the mouse model ofpseudomonas aeruginosa infection.
The pas-sive transfer of 0. 5ml mixture of McAb A4and D4 3 hors before Challenge with a
lethaldose or LD50 live P. aeruginosa (1× 108 or z0.25 × 108) results in a protection rate of
80~100%. The opsonophagocytic activity of theMcAb A4 and D4 was evaluated. The
rusultsshowed that the monoclonal antibodies pos-sessed potent opsonic activity which
enhancedphagocytosis to P. aeruginosa by human neu-trophil cell even in the absence of
comple-ment. Furthermore, using the misture ofMcAb as a therapeutic agent for burned
pa-tients with P. aeruginosa infection. A goodtherapeutic effect was observed.
