胰岛细胞经门静脉的肝内移植

Transplantation of islet cells from the portal vein into the liver

目的 以猪的胰岛移植探讨胰岛细胞移植后穿刺出血及肝内感染的发生。方法 采用高渗枸橼酸盐嘌吟溶液经过猪的腹主动脉进行原位灌注后 ,进行胰腺切取 ,采用胶原酶P消化胰腺 ,分离并纯化胰岛细胞 ,然后对猪胃左静脉穿刺后进行肝内胰岛细胞移植 ,对受者猪进行饲养 1周后 ,进行肝脏的病理学检查。结果 猪的胰岛细胞移植共进行 12例 ,穿刺部位出血占 5 0 % ,肝感染占 33. 3%。结论 胰岛细胞经门静脉的肝内移植过程中 ,门静脉出血高发和穿刺管道的直径有关 ,肝内感染率较高。

Objective To study the bleeding fromthe portal vein needle puncture site and intra-hepatic infection after islet cell transplantation in pigs. Methods Hypertonic citrate adenine solution was perfused into aorta. Pancrease was removed and digested with collagenase P. Islet cells were isolated and purified before intra-hepatic transplantation via puncture of the left gastric vein. Pathological examination of liver was performed one week later. Results Islets transplantation was performed in 12 pigs, The incidence of bleeding from the portal vein needle puncture site was 50% and the incidence of infection of liver was 33.3%. Conclusion High incidence of bleeding from the portal vein is related to diameter of the puncture needle the incidence of intra-hepatic infection is high.
