摘要
以毛泡桐(Paulowniatomentosa)无菌苗叶片和茎段为外植体,在附加NAA0.3mg·L-1或2,4 D0.3mg·L-1与6 BA5~20mg·L-1植物生长调节物质组合的MS培养基上进行体细胞胚胎发生及植株再生的研究.结果表明,毛泡桐叶片和茎段在不同植物生长调节物质组合的12种培养基上愈伤组织诱导率可达100%,但在2,4 D与6 BA植物生长调节物质组合的MS培养基上,叶片和茎段胚性愈伤组织诱导率为0,而在MS+NAA0.3mg·L-1+6 BA17mg·L-1培养基上,叶片和茎段的胚性愈伤组织诱导率分别达到88.3%和46.7%.胚性愈伤组织在该培养基上可发育为成熟的体细胞胚胎,进而成为完整植株.
Somatic embryogenesis and plantlet regeneration from leaves and nodal segments of Paulownia tomentosa were studied on MS media supplemented with NAA 0.3 mg·L^(-1) or 2,4-D 0.3 mg·L^(-1) and 6-BA 5~20 mg·L^(-1).The results showed that callus ratio of leaves and nodal segments was up to 100%. Embryogenic callus ratio of leaves and nodal segments on MS + NAA 0.3 mg·L^(-1) + 6-BA 17 mg·L^(-1) medium was 88.3% and 46.7% respectively, but the ratio on the media supplemented with 2,4-D was zero. Embryogenic calli could develop into mature somatic embryoes and plantlets.
