摘要
在Langendorff离体大鼠心脏模型,用含0.044g/LtritonX-100的改良K-H氏液经主动脉恒速逆灌大鼠心脏2min,造成去冠脉内皮功能的离体大鼠心脏实验模型。为证实该模型的可靠性,进行了有关试验的论证。结果显示:本方法可造成功能性无冠脉内皮而对血管平滑肌及心肌的生理功能无明显影响的离体大鼠心脏实验模型。本模型简便、稳定、可靠和实用。
The Langendorff rat hearts were perfused with a odified Krebs-Henseleit buffer(K-H-B)containing 0.044g/L triton X-100(end concentration)by cannulation of the aorta at a onstant flow(8ml·min^(-1)) for 2 min and then with a K-H-B without triton X-100 to establish a novel experimental model of removing endothelial cell function in coronary vessels.In the above-established model,prior to and after triton X-100 treatment:1)Car-dial function parameters(HR,CPP,LVSP,±dp/dt_max and so on)were measured and re-mained stable for 0 min;2)Vascular effects Induced by angiotensin Ⅰ(an endothelium-de-pendent vasoconstrictor)and Histamine(an endothelium-dependent vsasodialator)were abolished,whereas angiotension Ⅱ(an endothelium-independent vasoconstrictor)induced vasoconstrictlon remained unchaged,which showed that physiological functions of vascular smoothe muscle were not affected.Results showed that this novel isolated perfused rat heart model can achieve removal of conronary endothelial cells without altering physiological func-tion of vascular smooth muscle and myocardium.This model is a simple and convenient,sta-ble,reliable and practical method.
