多克隆抗体免疫荧光法检测大鼠卡氏肺孢子虫的研究 被引量:1

摘要 采用特异多克隆抗卡氏肺孢子虫囊壁抗体检测大鼠支气管肺泡灌洗液和肺组织中卡氏肺孢子虫,被荧光抗体标记的肺胞子虫呈圆形或卵圆形,直径约3~6μm,为特异的苹果绿荧光色的囊壁。受检标本经PronaseE溶解包裹在肺孢子虫包囊外的糖蛋白等物质,其检出效果优于不用PronaseE方法者,亦优于传统的Grocott和Giemsa染色法。本方法具有高度的敏感性和特异性,与白色念珠菌、新型隐球菌无交叉反应。 Pneumocystis carinii in the BAL and the lungs of rats were detected by special polyclonal antibodies against the wall of the Pneumocystis carinii cyst.P.carinii marked by the fluorescent-antibody were round or eggshaped,3—6 pm in diameter and had the special apple-green cyst wall.When P.carinii with glucoprotein outside the cyst were dissolved by the pronase E,the result was much better than that without using the pronase E.The method is superior to the conventional staining technique such as the Grocott and the Giemsa and has higher sensitivity,specificity,and no cross-reactivity with Candida albicans and Cryptococcus neoformans was found.
