摘要
目的 研究在E .coli中表达血管紧张素Ⅱ受体 1(angiotensinⅡreceptor 1,AT1)细胞外第二环肽段。方法 应用PCR技术 ,从大鼠基因组中扩增出目的基因 ,与表达载体 pGEX - 4T - 1重组 ,采用PCR、限制性内切酶酶切及DNA测序进行鉴定 ,将重组质粒转化到E .coliBL2 1(DE3)中进行表达。结果与结论 实验通过采用基因克隆技术构建并鉴定了AT1细胞外第二环的表达载体 ,并表达了相应的肽段 。
Objective To research the peptide corresponding to the second extracellular loop of angiotensin Ⅱ receptor 1(AT_1)expression in E.coli. Methods The target gene was obtained from rat genomic DNA by PCR,then recombined with the expression vector pGEX-4T-1 and identified using PCR, restricted endonuclease and DNA sequence analysis. Finally, the recombinant vector was expressed in E.coli BL21(DE3). Results The recombinant vector was constructed and identified using genetic cloning technology, and the target peptide was produced in E.coli BL21(DE3) in this experiment. Conclusion The experiment will establish the basis for studying the mechanism of autoantibody against the peptide.
