五倍子对兔眼结膜成纤维细胞的抑制作用（Ⅰ）

INHIBITORY EFFECT OF GALLA CHINESIS ON FIBROBLAST OF RABBIT OCULAR CONJUNCTIVA(Ⅰ)

报告了用五倍子提取液对体外培养的３～５代兔眼结膜成纤维细胞的抑制作用，结果显示五倍子提取液对成纤维细胞有显著抑制作用。五倍子终浓度２５μｇ／ｍｌ对成纤维细胞的抑制率为１７．２４％，与不加药的对照组比较差异显著（Ｐ＜０．０５），五倍子提取液终浓度越高对成纤维细胞的抑制率就越高，五倍子提取液终浓度７５～６００μｇ／ｍｌ对成纤维细胞的抑制率为３６．４８％～９７．８９％，与对照组比较差异非常显著（Ｐ＜０．００１）。细胞形态学观察结果可见五倍子对成纤维细胞具有明显的影响。与对照药物５－Ｆｕ及高三尖杉酯碱对成纤维细胞的抑制作用相等。３Ｈ－ＴｄＲ掺入试验提示五倍子抑制成纤维细胞的作用机理是抑制成纤维细胞的ＤＮＡ合成。

If subconjunctival fibroblast excessive proliferation after glaucoma filtering operation can block the humor aqueous filtering flux,then caused the operative failure.This paper reported the inhibitory effects of Galla Chinesis extractive solution on 3～5generation fibroblast of cell culture in vitro of rabbit ocular conjunctiva. The results showed that Galla Chinensis has significant inhibitory effects on fibroblast The inhibitory rate of 25μg/ml end concentration of Galla Chinesis extraction solution was 17.24%,has significant difference between Galla Chinesis group and without drug's contrary group(P<0.05).The end conentration of Galla Chinensis was more high and the inhibitory rate on fibroblast was more high.The inhibitory rate of 75μg/ml-600μg/ml of Galla Chinesis end conecntration was 36.48%-97.89%,compared with contrary group has remarkable significant differnce(P<0.001). Cell morphological observation showed that Galla Chinesis has obvious influence on fibroblast.Compared with contrary drug' s group,5-Fu and Homoharrangtonine, the inhibitory effects were equalize.The H-TdR test showed that mechanism of inhibitory effect of Galla Chinesis was inhibition DNA synthesising of fibroblast.
