蝎毒和蝎毒多肽对家兔血小板聚集的影响 被引量:2

摘要 目的 观察蝎毒多肽 (PSV)和蝎毒 (SV)对家兔血小板聚集的影响。方法 麻醉家兔 ,颈动脉取血制备富含血小板血浆 ,分别加入PSV和SV ,用CHRONO—LOG 5 4 0VS双通道血小板聚集仪测定二磷酸腺苷 (ADP)诱导的血小板聚集作用。结果 PSV对血小板聚集具有明显的抑制作用 ,且呈明显的量效关系 ,当PSV的浓度在 10 .0~ 15 .0mg/ml时 ,血小板聚集受抑制最为显著 ;SV对血小板聚集受其剂量及作用时间影响很大 ,但总的趋势为较低浓度时促进血小板聚集 ,较高浓度时则抑制血小板聚集。结论 PSV对血小板聚集起抑制作用 ,且呈明显的量效关系 ;SV对血小板聚集的影响在高浓度和低浓度时产生的效应相反 ,推测由于SV成分较为复杂 ,各成分之间互相作用机制还需深入研究。 Objective To observe the influences of PSV (peptide of scorpion venom) and SV (scorpian venom) on platelet aggregation in rabbit.Methods Arterial blood samples were drawn from the carotids of anesthetized rabbit, and then the platelet rich plasmas(PRP) were prepared. After adding PSV and SV to the PRP, the ADP-induced platelet aggregation was measured with the CHRONO-LOG540VS double channel multifunctional aggregation method.Results PSV was found to markedly inhibit the platelet aggregation in rabbit, and it showed a obvious dose-effect relationship. When the concentration of PSV was between 10.0~15.0 mg/ml,the platelet aggregation function was most markedly decreased; the anticoagulant effect of SV was markedly influenced by dose and effectible time, but low concentration of SV made advance at the platelet aggregation, otherwise high concentration restrained the platelet aggregation.Conclusion It is suggested that the PSV has an inhibitory effect on platelet aggregation, and it shows a dose-effect relationship. Anticoagulant effect of SV in high dose is opposited to low dose. It is supposed that the composition of SV is complicated, and the mutual function is not very clear.
