摘要
目的 对K5 6 2细胞向红系分化的中药血清药理学方法进行探讨。方法 以K5 6 2细胞为体外模型 ,采用联苯胺定性技术检测细胞内血红蛋白 ,比较不同条件下的含药血清诱导K5 6 2细胞前后的阳性率 ;测量波长为 4 14的A值。结果 10 %的含药血清培养诱导K5 6 2细胞向红系分化的作用最强 ;灭活血清对K5 6 2细胞向红系分化的诱导作用强于未灭活血清 ;大鼠qd连续 7d和bid连续 3d ,2种方法所得血清均以 10 %的浓度培养K5 6 2细胞 ,二者诱导其向红系分化的作用无明显差异 ;临床等效剂量ig所得血清比 2倍、4倍剂量要好。结论 含药血清的制备和含药血清在体外的合理应用对实验结果有很大的影响。
Aim To discuss the method in herbal serologic pharmacological by the inducement of erythoid differentiation in K562 cells.Methods K562 cells,used as a vitro model system,were stained with benzidine to assess hemoglobin(Hb) production in response to different treatment regiments of herbal medicinal mixture at varied concentration.Comparison of the percentage of benzidine-positive cells(BZ%)in different conditions was performed.Protein absorption at 414nm using a spectrophotometer were employed to determine the changes of Hb production in K562 cells.Results The effect of erythroid differentiation is the most powerful in regiments of 10% pastille serum than those in other densities.The function of erythroid differentiation in inactivated serum is better than that in activated serum.There are not significant differences in erytyroid differentiation by the both administration routes(2 times a day,continue 7 days and 1 time a day,continue 3 days).Conclusion The correct preparation and reasonable application of serum with medicine make very important influences on experimental results.
