摘要
目的 建立用气相色谱法同时测定石菖蒲挥发油中α 细辛醚和 β 细辛醚含量的方法。 方法 采用毛细管气相色谱法测定α 细辛醚和 β 细辛醚的含量。色谱柱 :HP - 35毛细管气相色谱柱 ;程序升温 :起始温度为 30℃ ,6℃ /min升至 2 2 0℃ ,保持5min ;进样口温度 :2 5 0℃ ;FID检测器温度 :32 0℃ ;进样量 :2 .0 μl;无分流进样 ;溶剂 :甲醇 ;载气 :氦气 ;柱头压 :5 .5× 10 4Pa ;内标物 :α 萘酚。结果 α 细辛醚的回归方程 :Y1=1.5 6× 10 -3 C1+4.12× 10 -3 (n =5 ,r =0 .9999) ;线性范围 :2 .0~ 2 0 .0 μg·ml-1;β 细辛醚的回归方程 :Y2 =0 .0 1889C2 +1.75 5× 10 -3 (n =5 ,r =0 .9999) ,线性范围 :2 0~ 10 0 μg·ml-1;α 细辛醚和 β 细辛醚的回收率分别为 99.6 %和 99.7% ,最低定量浓度分别为 2、2 0 μg·ml-1。结论 方法灵敏 ,定量准确 ,重复性好 ,适用于石菖蒲挥发油中α 细辛醚和 β 细辛醚的含量测定。
Aim To develop a method for determine α-Asarone and β-Asarone in volatile oil from Acorus.Tatarinowii Schott.Methods A capillary GC method was applied to determine the concentration of α-Asarone and β-Asarone.The GC system included:HP-35 column,Helium as carrier gas.Injection volume was 2.0μl with splitless inject mode.α-naphthol was used as interior standard substance.Results The linear equation of α-Asarone:Y 1=1.56×10 -3 C 1+4.12×10 -3 (n=5,r=0.999 9) was obtained in the range of 2.0~20.0μg·ml -1 (Y 1:ratio of the peak area of α-Asarone and of α-naphthol,C 1 :concentration of β-Asarone).The linear equation of β-Asarone:Y 2=0.018 89C 2+1.755×10 -3 (n=5,r=0.999 9) was obtained in the range of 20.0~100.0μg·ml -1 (Y 2:ratio of the peak area of β-Asarone and of α-naphthol,C 2 :concentration of β-Asarone).The recovery of α-Asarone and β-Asarone were 99.6% and 99.7% respectively.The limit of quantification 2~25μg·ml -1 of α-Asarone and β-Asarone were achieved respectively.Conclusion The method is accurate,sensitive and suitable.
