邻苯二酚紫共振光散射光谱法测定蛋白质的研究 被引量:1

摘要 基于人血清白蛋白 (HSA)对邻苯二酚紫的共振光散射的增强效应 ,拟定了一种测定HSA的新的共振光散射体系。在pH 2 5 6的Britton Robinson缓冲溶液中 ,邻苯二酚紫与HSA结合导致共振光散射增强 ,在λ =5. 15 0nm处共振光散射有最大的强度 ,并且共振光散射强度与HSA的浓度在 0. 0~ 2 0. 0 μg/mL范围内有良好的线性关系 ,检测限可达 0 . 18μg/mL。该方法简便、快速 ,用于人体血清样品的测定并与经典的考马斯亮蓝法比较 ,结果满意。 The method was based on the enhancement effect of the resonance light scattering(RLS) due to the combination of pyrocatechol violet with HSA.The RLS of pyrocatechol violet was greatly enhanced by HSA in the pH 2.56 Britton-Robinson buffer.The enhanced intensity of RLS at 515.0nm was proportional to the concentration of HSA in the range of 0.0~20.0μg/mL.The detection limit was 0.18μg/mL.The method was simple,rapid and has been applied to the determination of protein in human serum sample with satisfactory results.
