辛伐他汀对缺血再灌注损伤心肌保护机制的实验研究 被引量:2

摘要 目的 研究辛伐他汀对心肌缺血再灌注损伤的保护机制。方法 将健康雄性 SD大鼠随机分为假手术组、对照组和辛伐他汀组 ,辛伐他汀组于结扎冠脉前 12小时腹腔注射给药 ,假手术组、对照组注射等量生理盐水 ,八导生理记录仪记录缺血 1、30分钟及再灌注后 1、30分钟心功能 ,以图像分析软件计算缺血和坏死心肌面积。同时对缺血心肌行浸润 PMNs(多形核细胞 )计数。结果 缺血再灌注后与对照组比较 ,辛伐他汀组坏死区面积与缺血区面积之比减少 2 1% ,坏死区面积与左室面积之比减少 2 2 % (P均 <0 .0 5 ) ;左室内压上升段最大变化速率(+dp/ dtm ax)、左室内压下降段最大变化速率 (- dp/ dtmax)、心率 (HR)、心肌纤维缩短速率最大值 (Vmax)分别在再灌注 1分钟和 30分钟明显改善 (P均 <0 .0 5 ) ,缺血心肌 PMNs浸润明显减少。结论 辛伐他汀对心肌缺血再灌注损伤有保护作用。 Objective To investigate the effects of Simvastatin on ischemia-reperfusion injury in rat model.Methods SD rats were anethetized,and the left anterior descending coronary artery was ligated for 30minutes.After reperfusion for 30minutes,the ischemic and infarct size were determined.During the performances,the function of heart was recorded at different time.The myocardiac infiltrating PMNs number was counted.Results After LAD was ligated,+dp/dtmax、-dp/dtmax、HR、Vmax decreased and LEDP increased.Well the left ventricular systolic pressure had no change.+dp/dtmax、-dp/dtmax、HR、Vmax decreased further with reperfusion.Compared with control,+dp/dtmax、-dp/dtmax、HR、Vmax improved significantly in Simvastatin-group at 1minute and 30minutes,respectively(P<0.05).However,the systolic pressure of left ventricular had no change.At the same time Simvastatin-reduce infarction size significantly.The ratio of nec and aar decreased 21%(P<0.05).Meanwhile the ratio of nec and lv reduced 22%(P<0.05).PMNs infiltration number decreased in Simvastatin treated group.Conclusion Simvastatin can reduce myocardial infarction and improve contractile function caused by ischemia-reperfusion injury.
