延边黄牛卵母细胞激活及孤雌发育

摘要 探讨了提高延边黄牛卵母细胞激活率及孤雌发育率的适宜方法。从屠宰场回收的卵巢中采集未成熟的卵母细胞 ,置于体外成熟培养液中 ,体外成熟培养 2 0~ 2 2 h。将体外成熟的卵母细胞 ,分别用不同浓度的乙醇或 Ca- ionophore进行单一激活处理。试验结果表明 ,用 5 %、7%、9%、1 1 %及 1 3 %乙醇激活卵子 ,激活率分别为 4 1 .2 %、4 7.2 %、5 3 .1 %、3 9.7%及 2 3 .4 %,7%和 9%乙醇处理组的卵子激活率显著高于其他组 (P<0 .0 5 ) ,但二者之间差异不显著 (P>0 .0 5 )。用 2 .5、5 .0、7.5μm ol/L Ca- ionophore激活卵子 ,激活率分别为 2 9.3 %、5 1 .8%、4 5 .4 %,5 .0、7.5μmol/L Ca- ionophore处理组的卵子激活率显著高于 2 .5 μmol/L 组 (P<0 .0 5 )。用 9%乙醇或 5 .0 μm ol/L Ca- ionophore分别与 1 0 m g/Lcyclohexim ide(CH)组合激活卵子 ,卵子的激活率分别为 77.3 %和 76 .8%,比单独用 9%乙醇 (48.8%)或 5 .0μmol/LCa- ionophore(43 .2 %)的激活率显著提高 (P<0 .0 5 ) ,二者卵子的卵裂率和发育到 The purpose of this study was to determine the optimal conditions for parthenogenetic activation of in vitro matured Yanbian yellow cattle oocytes.Bovine follicular oocytes were aspirated from ovaries obtained from a local abattoir and matured in vitro for 20 to 22 h at 39℃,5% CO_2 environment in Medium 199 with 10% FBS and hormones.Matured oocytes were treated by various concentration of ethanol or Ca-ionophore alone and treated with 9% ethanol or 5.0 μmol/L Ca-ionophore combined with 10 mg/L cycloheximide.The activation rates were 41.2%,47.2%,53.1%,39.7% and 23.4%,respectively,following exposure for 5 min to 5%,7%,9%,11% and 13% ethanol.The percentage of oocytes activation were 29.3%,51.8% and 45.4%,respectively,after exposure for 5 min to 2.5,5.0 and 7.5 μmol/L Ca-ionophore.The rates of oocyte activation were 77.3% and 76.8% when oocytes were expoused to 9% ethanol or 5.0 μmol/L Ca-ionophore for 5 min and then cultured in 10 mg/L cycloheximide for 7 h,the corresponding activation values treated by 9% ethanol or 5.0 μmol/L Ca-ionophore alone were 48.8% and 43.2%.Parthenogenetic development was similar between 9% ethanol+10 mg/L cycloheximide and 5.0 μmol/L Ca-ionophore+10 mg/L cycloheximide.
