摘要
采用79对SSR引物分析了86个玉米杂交种的72份亲本自交系的遗传多态性,从中筛选出扩增带型清晰、稳定的50对引物用于构建86个杂交种的DNA指纹图谱数据库,进而确定10对核心引物和判别标准用于亲子鉴定研究。结合玉米SSR分子标记检测技术和单籽粒DNA快速提取方法,DNA指纹数据库和判别标准可有效地用于玉米杂交种的亲子鉴定。
The polymorphisms of 72 parental inbred lines from 86 maize hybrids were investigated by 79 pairs of SSR primers, and 50 primers with stable amplification profiles were selected to construct DNA fingerprinting database of 86 hybrids. The fingerprinting database and the testing procedure chosen were approached in parentage identification of maize hybrids. As SSR marker technique and DNA rapid extraction method established, the DNA fingerprinting database could effectively be employed to identify parentage relations among hybrids.
