摘要
目的:观察中药固本化痰健脑方对拟痴呆(AD)模型大鼠胆碱能神经系统功能的影响。方法:用鹅膏蕈氨酸建立大鼠AD模型,用改良Ellman法测定大脑组织乙酰胆碱酯酶(AchE)活性,用组织化学染色及图像分析技术测定AchE纤维密度,观察AchE在拟痴呆模型大鼠大脑海马和纹状体尾壳核内的不同分布及密度改变。结果:模型组能显著性增高脑AchE活性(P<0.01),空白组、阳性对照组、固本化痰健脑方高剂量组和低剂量组的AchE活性显著性低于模型组(P<0.01);在纹状体尾壳核CPU区和海马CA区,空白组、阳性对照组、固本化痰健脑方高剂量组和低剂量组与模型组相比均具有较高的积分光密度(P<0.01)。结论:固本化痰健脑方对鹅膏蕈氨酸所引起的脑内胆碱能神经元损伤具有一定的保护作用,其作用机制与抑制乙酰胆碱酯酶活性和提高Ach递质的量有关。另外,固本化痰健脑方高剂量较低剂量疗效佳。
Objective To observe the effects of Guben Huatan Jiannao Decoction (GHJD) on the Cholinergic system in rat model with Alzheimer's disease. Methods: We used injection of basal nucleus of Meynert (BNM) with ibotenic acid(IBO) to make AD model, We used the method of Ellman to assay the activity of acetylcholinesterase(AchE), and tissue chemistry method was used to detect the content of acetylcholine (Ach) in hippocampus and corpus striatum areas of brain. Results: The activity of AchE was higher in AD model rats than that of control group, piracetam treated, and the high-, low-treated group of GHJD(P<0.01); There was the remarkabe improvement on the number of Ach in the three medicine groups (P<0.01), and the activity of Ach in the high-treated group of GHJD was near to the level of control group (P<0.01). Conclusion: GHJD can improve the disability of learning and memory induced by ibotenic acid, and its effect maybe involve the activity of the acetylcholine system. What's more, the high dose of GHJD is better than that of the low one.
