摘要
以地高辛配基、光敏生物素及补骨脂素标记的N-rasDNA片段为探针,采用点渍法和DNA印渍法,比较了这三种非放射性标记探针的杂交灵敏度.在点渍法中,三者能检测到的低限DNA浓度分别为0.1μg·L-1,1μg·L-1和1μg·L-1.在DNA印渍法中,地高辛配基能检测到的低限DNA浓度亦为0.1μg·L-1.本文还讨论了影响上述三者灵敏度的有关因素.
Taking the N-rasDNA fragment labelled with Digoxigenin, Photobiotin and Psoralenas probes, and using both dot blotting and southern blotting, compared the hybridization sensitivity ofthree kinds of nonradioactively labelled probes. In the Dot blotting, the DNA concentration of minimun detectable amount of three 18belled compoundS iS 0. 1μg·L-1, 1μg·L-1 and 1μg·L-1 respectively. In the Southern blotting, the DNA concentration of minimun detectable amount of Digoxigenin is 0.1μg·L-1. The relative factors affecting the sensitivity of above three labelled compounds were also discussed.
