摘要
目的 :建立保健食品和果蔬中维生素C (VitC)含量的快速测定方法。方法 :样品用 5g/L草酸超声提取 ,采用高效液相色谱法 ,以 0 . 0 10mol/LNH4Ac -HAc缓冲溶液 (pH4 . 5 )作流动相 ,经C18色谱柱分离 ,用紫外检测器 ,在2 6 2nm波长下测定维生素C的吸光度值 ,并与标准系列比较 ,以保留时间定性 ,峰面积定量。结果 :本方法的线性范围为0 . 5 0~ 10 0 0 μg/ml,检出限为 5 0ng/ml,样品的加标回收率为 88 .5 7%~ 99. 12 % ,相对标准偏差为 1 .31~ 2. 82 %。 结论 :本方法灵敏度高 ,选择性好 ,操作简便、快速 ,5min即可完成定量检测 ,具有实用价值。
Objective:To establish a rapid method for the determin ation of vitami n C in healthy foods, fruits and vegetables by high performance liquid chromatog raphy.Methods:Vitamin C in samples was extracted by ultrasonic wi th 5.0 g/L ox alic acid. Vitamin C was chromatographically separated on a C18 column with a m o bile phase consisting of 0.01 mol/L ammonium acetate buffer solution (pH 4.5), a nd then detected at 262 nm by ultraviolet-visible detector.Results: The linear range of the proposed method was 0.5~1000 μg/ml.The detection limit of vita min C was 50 ng/ml. The average spiked recoveries of the samples were 88.57%~99. 12% with the relative standard deviation of 1.31%~2.82%. The determination of vi tami n C in samples could be finished in five minutes.Conclusion:The m ethod is simple,sensitive and applicable to the rapid determination of vitamin C in healthy foods,fruits and vegetables.
