己酮可可碱互穿网络凝胶微球制备及体外释放特性研究 被引量:3

摘要 目的 制备pH/温度双敏感互穿网络凝胶微球 ,考察微球对己酮可可碱的释放敏感特性。方法 以海藻酸盐为共聚模板 ,采用自由基水溶液聚合制备双敏感互穿网络凝胶微球 ,并考察药物体外释放对温度 ,缓冲液pH值敏感特性。结果 温度及缓冲液pH值对凝胶溶胀性能及药物的释放速率均具有显著影响。结论 所制凝胶微球具备pH/温度双敏特性 。 OBJECTIVE: To prepare thermo-and pH-sensitive interpenetrating network (IPN) hydrogel beads composed of calcium alginate and Poly(N- isopropylacrylamide) (PNIPAAm) hydrogels and investigate its controlled release characteristics with pentoxifylline as a model drug. METHODS: The gel bead was prepared by free-radical polymerization in aqueous solution using redox initiator in a simultaneous interpenetrating network process. The effects of temperature and pH on gel swelling performance and beads release characteristics in vitro were discussed. RESULTS: Temperature and pH appeared to significantly affect gel swelling ratio and the drug release rate. CONCLUSION: The IPN hydrogel beads containing pentoxifylline exhibited thermo-and pH-sensitive release characteristics in vitro. The drug release was the coupled action of diffusion and erosion mechanism. IPN matrix may be useful in the field of oral drug controlled delivery.
