PCR法检测乙型肝炎患者外周血单个核细胞中HBV　DNA

DETECTION OF HBVDNA IN PERIPHERAL BLOOD MONONUCLEAR CELLS OF PATIENTS WITH HEPATITIS B BY POLYMERASE CHAIN REACTION

采用ＰＣＲ法检测了７７例乙肝患者外周血单个核细胞（ＰＢＭＣ）中ＨＢＶＤＮＡ，２７例阳性，总阳性率为３５．１％。慢性活动性肝炎（９／２１，４２．９％），肝炎后肝硬化（９／１９，４７．４％）及重型肝炎（８／１６，５０．０％）患者ＰＢＭＣＨＢＶＤＮＡ阳性率明显高于慢迁肝（１／１４，７．１％）和急肝（０／７）患者。结果提示ＰＢＭＣ中ＨＢＶＤＮＡ的存在在乙肝发病机理中具有一定意义。

By means of polymerase chain reaction(PCR) technique, seventy- seven patients with various clinical types of hepatitis B were studied for the detection of hepatitis B virus (HBV) DNA in peripheral blood mononuclear cells (PBMCs).Twenty-seven cases were positive for PBMC- associated HBV DNA(35.1%).The positive rate of PBMCassociated HBV DNA in patients with chronic active hepatitis(9/21, 42.9%), posthepatitic cirrhosis(9/19,47.4%), and severe hepatitis(8/16,50.0%)was significantly higher than that in patients with chronic persistent hepatitis(1/14,7.1%)and acute hepatitis(0/7).These findings suggest that the existence of HBV DNA in PBMCs might play some roles in the pathogenesis of hepatitis B.
